Generally, any solid support in powder form that is chemically stable under the frit manufacturing conditions can be incorporated into a frit. Controlled pore glass (CPG) and polystyrene are common examples. What ultimately determines whether or not a support can be incorporated into a firit is the particular chemical modification that is present on the solid support. As an approved distributor of modified CPG (controlled pore glass) from AM Chemicals, we can supply you with several types of CPG Universal supports for DNA/RNA oligo synthesis. We find that some customers prefer to supply their own solid support. We are happy to work with either approach.
Yes, we can fill any size of plate with frits, including the standard 96, 384, 1536-well or custom plates. We can also prepare bulk frits of your preferred geometry, that you can load into a plate format, if that is your preference. We find that the scale factor of DNA purification frits is sometimes different from DNA synthesis frits due to the functioning capacity of the specific purification support. We are happy to share our experience with you and work together to develop a purification plate that meets your needs.
The exact answer to this question depends on the size of the frit, the bulk properties of the support and the specific application. Typically, our frits are 50% solids and 50% pores. Of the total solid volume in the frits, the maximum CPG loading is about 40 - 60 vol%.
No, there is no minimum loading level. However, if you are thinking to reduce the loading level of solid support in your frit in order to conserve synthesis reagents, you should also consider reducing the overall frit volume because extra bed volume absorbs reagents and makes inefficient use of those expensive reagents. We can help you identify the best frit size and loading of solid support for your application.
Depending on the physical properties of the solid support, we may be able to manufacture frits having a dimension as small as 0.5 mm. Please contact us if you are interested in miniaturizing your frits. We may have standard products that meet your needs already.
